1415A Permeabilized Fiber ATPase System
The 1415A Permeabilized Fiber ATPase System accurately measures various characteristics of fibers across a wide range of applications and tests. Conducting force-pCa experiments combined with energy metabolism measurements becomes exceptionally easy with our auto-indexing bath plate. Simply preset the calcium concentrations and activation/relaxation sequences—the 1415A system will take care of the rest automatically.
The system comes equipped with a temperature-controlled device, which includes an XYZ micromanipulator with an integrated mounting base. This is compatible with our high-speed length controller and force transducer.
The bath controller features dedicated software and a built-in programmable motion control sequencer, enabling automatic bath transitions for the fiber under study. An optional camera- or laser-based sarcomere length (SL) measurement system is available for setting the initial resting SL of skeletal and cardiac tissues.
An additional ATPase cuvette for measuring ATP consumption can be easily connected to our optical instruments. It excites the fluorescent dye in the solution using UV light and quantifies NADH absorbance in the bath using a photodetector. The system also includes a high-speed length controller, precision force transducer, data acquisition hardware, and our unique real-time Linux control and analysis software.
Our dedicated software includes a comprehensive experimental protocol library, simplifying the process and making the measurement of force and length straightforward. When combined with the optional HVSL sarcomere length measurement system, researchers can simultaneously control and measure length, force, and sarcomere length. A dedicated software module also enables precise ADP injection and provides graphical tools to measure the consumption rate.
These advanced features allow researchers to fully characterize permeabilized fibers and perform all standard tests, including force-pCa, kTr, ATPase activity, length-tension, force-velocity, and cross-bridge cycling kinetics.
The Aurora Scientific Permeabilized Fiber ATPase System is built using corrosion-resistant materials and can be easily mounted on a benchtop or inverted microscope, facilitating both basic observation and complex imaging.
Features
Test single permeabilized fibers, fiber bundles, and cardiac tissue
Temperature-controlled 10-well plate with removable ATPase cuvette for energy metabolism measurements
Measure force-pCa, kTr, ATPase activity, stiffness, length-tension, and force-velocity relationships
Software-controlled ADP injection
Includes UV light source and photodetector for NADH absorbance measurement
Built-in laser-based sarcomere length (SL) detection; compatible with benchtop or inverted microscopes
Automated Sample Bath Transfer
Thanks to our auto-indexing bath plate, conducting force-pCa experiments is simple and efficient. Simply preset calcium concentrations and activation/relaxation sequences, and the 1415A system will automatically handle the rest.
Powerful Standard Protocol Software
The software’s protocol library includes a variety of experiments designed for single fiber research, along with mini-applications for adjusting system settings. Powerful preset functions also allow you to add custom protocols, enabling efficient operation across multiple lab users.
Integrated Sarcomere Length Control
When the 1415A is combined with our 902A laser sarcomere length device or 901D video-based sarcomere length software, setting the resting sarcomere length becomes straightforward. The high-speed camera synchronizes with data acquisition software, enabling real-time, simultaneous collection of force, fiber length, and sarcomere length, all saved in one time-synced data file.
Built-in Optical System
The ATPase cuvette connects easily to our optical instruments. UV light excites the fluorescent dye in solution, and a photodetector quantifies NADH absorbance in the bath.
